Arachadonic Acid/Ceramide-Based Screens for Colorectal Cancer Chemopreventive Agents
Nonsteroidal antiinflammatory drugs (NSAIDS) can inhibit colorectal tumorigenesis and are one of the few agents known to be useful for the chemoprevention of neoplasia. It is shown that NSAID treatment of colon tumor cells induces apoptosis and a dramatic increase in ceramide, a known mediator of apoptosis. The ceramide increase is likely to be mediated by the elevated arachidonic acid resulting form cyclooxygenase inhibition, which in turn stimulates sphingomyelinase. These results have significant implications for understanding and improving colon cancer chemoprevention.
