Fluorogenic Substrates for Protein Phosphatases and Assays Incorporating the Substrates

Provided is a class of fluorogenic protein phosphatase substrates. These fluorogenic substrates comprises a carbamate moiety having a carbamate functional group other than hydrogen bonded to the carbamate nitrogen, a fluorophore containing a fluorophore functional group able to covalently bond to the oxygen of the carbamate moiety, and a peptide sequence bonded at its N-terminus to the carbon of the carbamate moiety and composed of one or more amino acids in which the N-terminal amino acid is either phospho-serine or phospho-threonine. The substrates may be employed in assays to identify inhibitors of protein phosphatases that are implicated in human diseases.
