Solvent recovery - Gathering in formation regarding the solvent i.e Methylene Chloride (MC) from the suppliers and other online resources as the problem with simple distillation was that the recycling result in drop of yield of ampicillin tri-hydrate / amoxicillin tri-hydrate. After going through the literature i found out that before distillation we need to adjust the pH<7 and washing the solvent with water to remove other low boiling components. Based on these hypothesis implemented solvent recovery program and was successful getting same yield profile as with fresh solvent. a great deal of cost saving.
In case of an extra peak in product monograph i perform root cause analysis on the manufacturing process and evaluated all the parameters and as i go through the monograph of the intermediates used in synthesis find out that the extra peak was in the range of the basic intermediate i.e Isoxamine (AMI), performing an process audit was able identify that the floor the chemist were adding Acetyl-chloride for acylation reaction at very temperature - resulting the dissolved AMI recrystallizing and resulting incomplete reaction and remaining/un-reacted AMI coming out with product and giving extra peak in the product monograph.
