Multi-photon Microscopy Via Air Interface Objective Lens

The present disclosure provides an optical system for depth-resolved imaging of a specimen such as living tissue in vitro (e.g., brain tissue). The optical system comprises a confocal and multi-photon microscope having an objective with adjustable glass correction and a gas between the objective lens and a specimen. The optical system also comprises an incubation chamber having a first housing with a glass aperture and a second housing enclosed by the first housing, said second housing having a glass aperture. The specimen is positioned within the second housing. The present disclosure also provides methods of using the optical system.
