Rapid Multiplex Gene and mRNA Detection in Picoliter Droplets by Molecular Beacons

Background: Breast cancer is a world wide public health concern and the estimated death toll in the U.S. in 2001 was 20,000 and it also ranked first as the cause of cancer death for women between the ages of 20 to 59. People carrying a mutation in the BRCA1and BRCA2 genes are prone to breast cancer and ovarian cancer. The conventional DNA detection method which involves a heterogeneous solid-liquid hybridization process requires probe immobilization. Stringent rinsing to remove specific DNA bonding is required and the whole process is time consuming and requires a large number of samples. Technology: A new technique using molecular beacons (MBs) for the construction of probes that are useful for real-time detection of nucleic acids has been shown. MB's are synthesized single stranded nucleic acid molecules that are constructed by stem and loop structures. The loop portion contains sequences complementary to the target single strand DNA and the stem portion is formed by annealing two complimentary arm sequences that are not related to the target single strand DNA. MB's have outstanding capability for the selective detection of single nucleotide polymorphism. This invention describes a novel picoliter droplet system that is a fast and simple gene detection platform. Application: Gene detection, breast cancer detection analysis, pathogen detection, virus detection, protein detection and antibody detection
