"Teabag" Processing: an Automated, High Throughput Method for Affinity Purification

Summary A high throughput system for protein purification has been developed, whereby a “tea-bag” structure is used to house microspheres for targeted protein binding and elution. The microspheres are compatible with any solid-phase protein purification, which includes coupling of Ni++/Histidine, Glutathione/Glutathione-S-transferase, avidin/biotin, and immunoprecipitation (beads with either protein A or G). From an efficacy perspective, the system has demonstrated equivalent purified protein yields when compared with manual methods, and product development can be scaled-up quickly for commercialization. Moreover, consumable plates (96 wells) or pipette tips may be deployed across a broad range of automation levels, including manual handling, custom workstations, or automated liquid handling robots.

Applications The need for high throughput protein purification will facilitate a variety of applications including in vitro study of protein-protein interactions; study of protein complexes; high throughput screening of small molecule ligands against protein targets; high throughput structural genomics (protein structure ia NMR and/or X-ray crystallography).
