Facile, PCR Free Detection of DNA with Single Nucleotide Discrimination

Abstract (Set) Single nucleotide modifications in DNA have important affects on gene expression. This results in significant phenotypic effects. Mutations in genes can also result in the development of diseases, including cancer. The ability to rapidly analyze DNA sequence is useful for correlating mutations with gene expression, and cellular properties. JHU scientists have developed a facile, sensitive method for detecting single nucleotide changes in regions of interest in DNA. Description (Set) ? Does not require PCR amplification of the target; ? More sensitive than other methods that do not utilize PCR: detects as little as 250 fmol of target; ? Uses common ELISA like techniques for detection; ? Requires minimal technical expertise and utilizes common, safe reagents; ? Compatible with experiments carried out in cells than sensitive nanoparticle based methods.
