Polymerase-inhibiting virustatic agent - extracted from root of Cochlospermum angolense as carotenoid, effective against HIV reverse transcriptase, influenza replicase, etc.

The inhibitor contains a compound (I) from the root of Cochlospermum angolense, with appropriate carriers and adjuvants. (I) is a carotinoid drug extract, obtained with water or alcohol from the finely powdered root. The extract is used in crystalline or amorphous form in tablets or capsules, or as drops. Dose is 100-200 micro-g/ml per unit, corresponding to 100-200 mg/kg body weight per day. USE/ADVANTAGE - The inhibitor is useful in the treatment of virus infections. It inhibits HIV reverse transcriptase, influenza replicase and alpha- and beta-DNA polymerase, calf thymus and E. coli RNA poilymerase. In an example, 800g ground C. angolense root was defatted with 600ml petroleum ether and extracted with 600ml CHCl3 in a Soxhlet process. 2g of red-brown extract was obtained by evaporation of the solvent. The extract was chromatographed on a Sephadex LH20 gel with MeOH as eluant. The orange-red extract produced a red crystalline powder by evaporation, with average yield of 10%.
