K. Srinivas Sashidhar's Problem Solving Experience:
- As a research scientist i have worked in the molecular biology area, I was involved in the synthesis of a chromosome of the artificial synthetic microbial organism(Organism name is confidential). We were given the task for designing the oligos for the segment of chromosome of the organism. We had found that after amplification some gaps were created and some essential genes may get deleted, so I suggested of using an overlapping sequence with essential genes in the gaps to be inserted, so that, the unnecessary mutations can be minimized when integrating it into the organism.
- I have also been part of a development team, for invitro abalysis of a generic drug nad optimization in various acidic and basic media, in order to improve its dissolution invivo. The confirmation of whether, it will act invivo was a problem, I had correlated with the invivo profile using blood samples and analysis was done using HPLC. I had established an a invitro-invivo correlation of the drug using a mathematical model, which reduces the no. of expensive bio-equivalence tests invivo. This tool also helps in further dissolution improvements. The optimization was successful and correlation was found that the drug was active in acidic state.
