Ali Rafati's Problem Solving Experience:

  1. 1. Denaturation: The DNA sample is heated to around 95°C to separate the double-stranded DNA into single strands.

    2. Annealing: The sample is cooled to around 50-65°C, allowing the primers to bind to the specific DNA sequences flanking the target region.

    3. Extension: The temperature is raised to around 72°C, and a heat-stable DNA polymerase enzyme synthesizes new DNA strands complementary to the single-stranded DNA templates, using the primers as starting points.

    4. Repeat: Steps 1-3 are repeated for multiple cycles (usually 20-40), resulting in exponential amplification of the target DNA sequence.
