Identification of Protein Ubiquitination Sites
Summary Protein ubiquitination is the most common of all postranslational modifications, and is a key process in the pathway of protein degradation. The present invention describes a method of identify protein ubiquitination sites. The process entails obtaining ubiquitinated polypeptides, digesting with proteases, determining the presence of an isopeptide bond in a test peptide by mass spectroscopy. The test peptide being evaluated can be ioinized and/or fragmented prior to the determining step.
Applications Ubiquination is a fundamental component in the degradation of proteins and is a key point of regulation in normal homeostasis and disease processes. Also, inhibition of the biquitin/proteosome pathway has led to development of a successful cancer drug, Velcade.
