Facile Modification of Collagen Directed by Synthetic Peptides

mechanical properties. Such modification may allow attachment of bioactive agent that can bring in designed bioactivity to natural collagen. Most of the techniques practiced today for collagen modification involves simple "blending method" which results in weak attachment of the target molecules or "chemical modification" that requires relatively harsh reaction conditions that are inapplicable to modifying collagens in living tissues. JHU researchers have developed a novel, simple approach for modifying natural collagen with minimum biochemical and structural perturbation of the native collagen. This technology provides for synthetic peptides that can interact specifically with native collagen. In addition, JHU researchers have discovered several design, synthesis and collagen modification procedures for a series of synthetic conjugates of peptide-fluorescence molecules and peptide-PEG molecules. Description (Set) Proposed Use (Set) This technology may be used to modify different forms of collagen or it can be used to deliver and attach therapeutic agents to collagens in the living tissue. For example, it may be used to incorporate therapeutic (antimicrobial or anti-inflammatory) agents to hemostatic collagen sponges or protective collagen-based corneal shields. It may also be developed as a research tool for tissue engineering matrix that is endowed with tailor-designed bioactivity.
