Generation of Isogenic Lines Differing Only in MLH1 Status

JHU researchers have created matched cell lines which are isogenic, differing only in the MLH1 alleles. One line has two defective alleles, the other has one defective allele and one normal (wild-type) allele, which was generated by targeted homologous recombination ("knock-in"). One wild-type allele is sufficient to generate normal mismatch repair. Description (Set) Proposed Use (Set) The cell lines can be used to test for compounds that are specifically toxic for cells lacking mismatch repair activity.
