Molecular Insights into Breast Cancer Prevention and Therapeutics Involving Novel Genistein Analogs

Abstract (Set) Cancer is the second leading cause of death in the U.S. and claims the lives of around 555,000 Americans each year. An estimated 1.3 million new cases of cancer are reported in the U.S. each year, of which breast cancer accounts for 15.3 percent. In Asia, an area with high soya isoflavone intake, breast cancer rates are lower than in the western countries. The potential estrogenic properties of genistein and other isoflavones have raised some concerns about their use in women at risk for breast cancer. JHU scientists propose a structure-based molecular modeling design approach to synthesize genistein analogs with idea to preserve genistein anti-tumor activity while eliminating its estrogen-like effects. In vitro characterization these new compounds in two breast cancer cell lines (MCF7 and T47D) showed that these derivatives exhibited improved anti-proliferative effects compared to genistein. Description (Set) Proposed Use (Set) New genistein analogs with improved anti-proliferated effects compared to genistein can be better candidates for breast cancer therapy.
