Development of Peptide Antibody Depletion (PAD) and its Application to Mass Spectrometry Sample Preparation

One of the major limitations of mass spectrometry is obtaining mass information on proteins in the presence of contaminating materials or other high abundant proteins that, when present in the sample, suppress the observation of proteins of interest. JHU scientists have developed a novel application called Peptide Affinity Depletion (PAD) which specifically targets the high abundant, well ionizing proteins and/or their resulting peptides/fragments and removes them prior to the ionization procedure. The sample preparation procedure improves the detection of proteins of interest during mass determination. An alternative application of this technology is the capture of specific highly ionizing peptides for the use as diagnostic markers. In this case, the diagnostic peptide of interest is captured and all other peptides depleted, vastly increasing the sensitivity of detection. • remove major abundant contaminants in mass spectrometry assays;
• enable the creation of an antibody library used in affinity products, sample processing instrumentation, and pre-columns for HPLC and nano-HPLC instrumentation directly coupled to mass spectrometers;
• diagnose certain conditions in patients;
