Treatment for keratinizing dermatological disorders by reduction in keratin expression

Compounds and a method for screening for a compound to reduce the expression of keratin proteins in keratinizing dermatological disorders are disclosed. Preferably keratin expression is reduced by the upstream targeting of the transcription factor stat1 using statins (HMG CoA inhibitors), such as Compactin, Mevastatin, Simvastatin, Atorvastatin, Cerivastatin, Fluvastatin, Lovastatin, Pitavastatin, Pravastatin or Rosuvastatin. Alternatively keratin expression may be reduced using RNAi strategies. Preferably the compounds are for the treatment of atopic dermatitis (eczema), ichythyosis vulgaris, epidermolysis bullosa simplex and particularly pachyonychia congenita.
